4Moving Biotech has completed enrollment in INFLAM MOTION, an international Phase 2a proof-of-concept study evaluating 4P004, an investigational intra-articular GLP-1 receptor agonist the company claims is first-in-class and designed to address inflammatory mechanisms underlying symptoms and disease progression in knee osteoarthritis.
The randomized, double-blind, placebo-controlled trial enrolled 129 patients across Europe, the United States and Canada with symptomatic knee osteoarthritis and contrast-enhanced magnetic resonance imaging confirmed synovitis.
Participants were randomized to a single 2 mg intra-articular injection of 4P004 or placebo and will be followed for three months. The primary endpoint is reduction in knee pain at Week 4; key secondary endpoints include changes in synovial tissue thickness and overall synovitis burden assessed by CE-MRI, knee pain at Week 12, imaging biomarkers and circulating biomarkers associated with osteoarthritis progression.
Patient follow-up is ongoing and topline results are expected by the end of December 2026.
According to the company, the data will inform patient selection, endpoint strategy and the design of a subsequent Phase 2b study and regulatory interactions.
