Alector confirmed it will discontinue the Phase 2 PROGRESS-AD trial of Nivisnebart (AL101/GSK4527226) in individuals with early Alzheimer’s disease after a pre-specified futility analysis by an independent data monitoring committee concluded the study was unlikely to meet its primary endpoint of slowing disease progression at completion.
Alector and GSK co-developed AL101, and the companies will work with investigators to inform trial participants; full results will be shared at a future medical meeting. AL101 is an investigational monoclonal antibody designed to block and downregulate the sortilin receptor, thereby elevating progranulin levels in the brain, a mechanism the company describes as linked to lysosomal function and neuronal survival.
Alector restated its focus on advancing its pipeline, enabled by the Alector Brain Carrier platform, the company claims, including AL037/AL137, an ABC-enabled anti-amyloid beta program targeting an IND submission in Q1 2027; AL050, an engineered glucocerebrosidase enzyme replacement therapy targeting an IND in 2027; ABC-enabled siRNA programs AL064/AL164 for tau and other programs ADP062-ABC and ADP065-ABC toward lead selection.
Alector is headquartered in South San Francisco and trades on Nasdaq under ALEC.
