Arvinas announced that the first participant has been dosed in its Phase 1/2 clinical trial of ARV-6723, an investigational oral PROTAC designed to degrade hematopoietic progenitor kinase 1 in adults with advanced solid tumors.
The ARV-6723-101 trial is a global, multicenter, first-in-human study designed to assess safety, pharmacokinetics, pharmacodynamics, and preliminary antitumor activity of orally administered ARV-6723 as monotherapy and in combination with pembrolizumab, and the initial monotherapy cohort is enrolling patients who previously received an immune checkpoint inhibitor and have no suitable standard treatment options.
Arvinas positions ARV-6723 as its first immuno-oncology clinical candidate and the first HPK1 PROTAC degrader to enter the clinic in the United States, and the company claims preclinical data showed potent and selective HPK1 degradation, enhanced immune activity, and antitumor activity as a single agent and with an anti-PD-1 therapy, including meaningful single-agent activity in seven models where an HPK1 inhibitor or anti-PD-1 therapy showed no benefit.
