Innovent Biologics has dosed the first patient in TriadicMM-1 (NCT07623798), a pivotal, multicenter, randomized, controlled, open-label Phase 3 study in China evaluating IBI3003, the firm’s tri-specific antibody targeting GPRC5D, BCMA and CD3, for second- to fifth-line relapsed or refractory multiple myeloma.
The trial will compare IBI3003 to the investigator’s choice of pomalidomide, bortezomib and dexamethasone (PVd) or daratumumab, pomalidomide and dexamethasone (DPd), with progression-free survival assessed by an independent review committee as the primary endpoint.
The company claims IBI3003, built on its Sanbody platform, showed superior antitumor activity in preclinical models versus marketed bispecific benchmarks and has progressed from Phase I/II studies in China, Australia and the U.S. into this registrational stage.
Data presented at the American Society of Hematology on 7 December 2025 reported an overall response rate of 83.3% among patients dosed at or above 120 μg/kg (n=24) in an early cohort and described predominantly Grade 1–2 cytokine release syndrome and mostly manageable treatment-emergent adverse events. IBI3003 also received U.S. FDA Fast Track designation earlier this year.
