Jupiter Neurosciences starts enrolling patients in a mid-stage study for JOTROL, an oral candidate targeting Parkinson’s at its roots.

Targeting Parkinson’s at its roots sounds straightforward in theory, but in practice, it has defeated scientists for decades.

Most treatments available today help patients regain some control over movement, but underneath those symptoms, the disease continues its silent progression. Brain cells keep deteriorating, inflammation lingers, energy production inside cells begins to falter.

That is the space Jupiter Neurosciences is now stepping into with the launch of its Phase 2a RESET trial for JOTROL, an investigational oral therapy for Parkinson’s disease. The company announced that patient enrollment is officially underway, with first dosing expected soon [1].

Increasingly, Parkinson’s research is borrowing ideas from longevity science – not just treating what patients feel, but trying to understand why vulnerable brain cells age, weaken and eventually fail in the first place.

Parkinson’s affects around 1.1 million people in the United States, with roughly 90,000 new diagnoses every year. Despite years of breakthroughs in neuroscience, there are still no approved therapies proven to slow or stop the disease itself. That reality has frustrated researchers for years because Parkinson’s is no longer viewed simply as a disorder of dopamine loss. Scientists now see a wider systems problem unfolding inside the brain.

Inflammation remains switched on for too long. Cells struggle to manage oxidative stress, essentially the biological wear-and-tear created during normal metabolism. Mitochondria, the tiny structures that generate energy inside cells, begin malfunctioning. Say neurons start behaving like cities running on unstable power grids. They can function for a while under pressure, but eventually outages begin appearing everywhere.

That growing understanding has reshaped the field. Rather than focusing solely on replacing dopamine, researchers are increasingly looking upstream at the aging-related processes creating the damage in the first place.

That is also where Jupiter’s investigational therapy enters the picture. JOTROL is based on trans-resveratrol, a compound that has floated around longevity conversations for years. It gained attention because laboratory research suggested it may influence pathways linked to inflammation, cellular stress and mitochondrial function.

The problem was never curiosity, but delivery. Standard resveratrol is notoriously difficult for the body to absorb efficiently. Much of it breaks down before enough can circulate meaningfully – especially in the brain, where therapies must cross additional biological barriers.

Jupiter says its micellar formulation was designed to improve that process. In earlier Phase 1 testing involving 24 healthy volunteers, the company reported roughly nine times higher bioavailability compared to conventional resveratrol. Researchers also detected measurable levels of the drug in cerebrospinal fluid, an important sign that the compound may actually be reaching the central nervous system rather than simply passing through the bloodstream. That does not prove effectiveness, but in neurodegenerative disease research, getting a compound into the brain consistently is already a significant hurdle cleared.

“Despite advances in symptomatic management, there remains a significant unmet need for therapies that may address the underlying biological drivers of Parkinson’s disease,” said Dr Charbel Moussa, MBBS, Professor of Neurology and Director of the Translational Therapeutics Program at Georgetown University Medical Center and Coordinating Principal Investigator of the RESET study.

Moussa expressed confidence in the therapeutic potential of JOTROL. He noted that the drug’s superior central nervous system exposure and unique mechanism of action distinguish it as a promising candidate for treating neurodegenerative conditions rooted in oxidative stress, mitochondrial failure and persistent neuroinflammation.

The RESET study will enroll 30 patients across three clinical centers connected to Georgetown University and MedStar Health. Over 12 weeks, researchers will evaluate safety, tolerability and biological markers tied to inflammation and cellular energy production.

Neurodegenerative diseases remain among the hardest areas in medicine to crack because the brain rarely offers quick answers. Changes unfold slowly, symptoms overlap and proving that a therapy truly alters disease progression can take years.

Still, investors and researchers continue pouring resources into the field because the need is impossible to ignore. Parkinson’s carries an estimated annual economic burden of more than $82 billion in the US alone.

“With enrollment now underway in the Phase 2a RESET trial, we have reached a critical operational milestone. We look forward to generating the clinical data needed to evaluate JOTROL’s potential and to serve patients, providers, and the broader scientific community,” said Christer Rosén, CEO of Jupiter Neurosciences.

For Jupiter itself, the trial represents more than a commercial milestone. Alison Silva, President and Chief Business Officer at the company, described the launch as the culmination of ten years of research and development work.

Results from the trial are expected in the first half of 2027.

Whether JOTROL succeeds remains uncertain. Parkinson’s research has humbled some of the biggest names in biotech and pharma. However, what’s happening across neuroscience recently feels increasingly difficult to ignore. Medicine may finally be moving closer to what researchers have been chasing all along: targeting Parkinson’s at its roots.

Longevity level(s):

  • Level 3: Longevity biohacking
Companies that manufacture ingredients and dietary supplements, or produce equipment such as red light therapy, cryonic chambers, and enhanced exercise equipment to maintain the metabolism and replace cellular function lost with the progression of age.
  • Level 6: Aging disease prevention therapeutics
Therapeutic interventions that control aging drivers to prevent aging diseases from occurring, like the current wave of GLP-1 agonists that prevent type 2 diabetes through metabolic interventions that also are proving effective in mitigating cardiovascular and neurological diseases.

Overview

Jupiter Neurosciences is a clinical-stage biopharmaceutical company developing a proprietary resveratrol delivery platform to address neuroinflammatory disorders and rare neurological diseases. The company's lead therapeutic candidate, JOTROL, utilizes patented micellar technology enabling oral administration of resveratrol at therapeutic levels without gastrointestinal side effects, with applications in Alzheimer's disease, Parkinson's disease, and rare conditions including mucopolysaccharidosis type I, Friedreich's ataxia, and MELAS. The company operates a dual-track strategy combining clinical development of JOTROL with commercialization of Nugevia, a consumer longevity supplement line, positioning itself at the intersection of precision therapeutics and healthy aging.
