Rznomics Inc., a clinical-stage biopharmaceutical based in Seongnam, South Korea, announced that research published online on 4 September in Nucleic Acids Research describes optimized methods for in vitro production of circular RNA.
The peer-reviewed paper details systematic target-site selection and P1 construct engineering within an end-to-end self-targeting and splicing workflow and reports up to two-fold higher self-circularization efficiency for long genes of about 8 kilonucleotides compared with the permuted intron–exon method.
According to the company, these optimizations improve the efficiency of its proprietary Self-Targeting and Splicing (STS) technology and could enable efficient circRNA production across a wider range of RNA lengths for vaccines and protein therapeutics.
The paper and the company note that the STS approach generates circRNA without extraneous sequences, which the authors say may avoid sequence-related immune responses and aid scalable manufacturing.
Rznomics is presented in the announcement as pursuing broader therapeutic applications of the platform while continuing its development programs in RNA-based gene therapies.
